dc.description.abstract Introduction: Oxidative stress plays a well-known role in acute ischemic stroke (AIS). AIS leads to an increase in free radicals and reactive oxygen species in tissues. While the exact mechanism is uncertain, oxidative stress has an important role in the ischemic pathway and may contribute to the deterioration. The aim of this study is to investigate the dynamic thiol/disulphide homoeostasis to evaluate the oxidative process in AIS with and without large-artery atherosclerosis. Materials and methods: Total thiol level (TTL), Native thiol level (NTL) and disulphide ratios of 30 patients with acute ischemic stroke and internal carotid artery (ICA) stenosis and 30 patients without ICA stenosis were compared between the two groups and 30 healthy control subjects. The calorimetric measurement and duplex formation methods used in the present study to measure dynamic thiol/disulphide homoeostasis have been developed in 2014. These new methods are simple, reliable and sensitive of high linearity and repeatability. Results: There was no difference in TTL, NTL and disulphide values between the groups with and without ICA stenosis (p>0.05). TTL, NTL and disulphide values of both groups were significantly higher than the control group (p<0.05). Conclusion: High disulfide value may be a marker of acute ischemic stroke independent of atherosclerosis.
