Bitkiler Antik çağdan beri halk tarafından yaraların ve enfeksiyonların giderilmesinde ayrıca hastalıkların tedavisinde kullanılır. Bu tedavi etme özellikleri insan vücudunda fizyolojik etki yaratan bazı kimyasal maddeleri bünyelerinde barındırmasından gelmektedir. Bitkilere tedavi edici özellik veren bu kimyasallardan bazıları alkaloidler, saponinler, tanenler, flavonoidler, fenolik bileşiklerdir. Bitkiler üzerine yapılan çalışmalar arttıkça bitkilerin faydaları daha çok gün yüzüne çıkmıştır. Bitkilerin içeriğinde bulunan önemli bileşenler bitki tüketimini arttırmakta ve alternatif tüketim yolları ortaya çıkarmaktadır. Bu tüketim yollarından en çok kullanılanlardan biride bitki içerikli detoks sularıdır. Detoks vücudumuzda bulunan toksinlerin giderilmesi ve nötralize edilmesidir bunun belli bir süreci vardır. Bitkiler bu sürece yardımcı olur. Çalışmamızda çeşitli bitkiler ile hazırlanan detoks sularının aktiviteleri araştırılmıştır. Yapılan çalışmada daha önce biyolojik özellikleri çalışılmış olan bazı bitkilerden hazırlanan karışımların toplam fenolik madde miktarı, antioksidan ve antimikrobiyal özellikleri araştırılmıştır. Çalışmada Malus domestica (Elma), Zingiber officinale (zencefil), Camellia sinensis (yeşil çay), Cinnamomum (tarçın), Citrus × limon (limon), Petroselinum crispus (maydanoz), Cucumis sativus (salatalık), Persea americana Mill.(avokado), Spinacia oleracea (ıspanak) ve bu bitkilerin birbiri ile kombine edilerek hazırlanan elma-tarçın, limon-zencefil, limon-yeşil çay, maydanoz- limon, salatalık-limon, salatalık-maydanoz, avokado-limon, maydanoz-salatalık- limon etanol ekstraktların B. subtilis ATCC 6633, S. aureus ATCC 29213, E. faecalis ATCC 29212, E. coli ATCC 25922, S. epidermidis ATCC 12228, S. abony NCTC 6017, S. typhimurium ATCC 14028 suşlarına karşı disk difüzyon yöntemi ile antimikrobiyal özellikleri araştırılmıştır. DPPH yöntemi kullanılarak antioksidan etkileri ve Folin-Ciocalteu reaktifi ile toplam fenolik madde miktarları belirlenmiştir.İlk olarak aktar ve pazardan alınan meyve ve sebzeler yıkanarak ılık su içinde demlenmeye bırakılmıştır. Yaklaşık olarak 1 saat kadar demlenerek hazırlanan detoks suları huni ve filtre kağıdı ile süzülerek uçurulmuştur.Uçurulan detoks suyu içerisinde kalan maddeler uygun ölçüde hazırlanan etanol konsantrasyonu ile çözdürülerek bitki detoks sularınıdan ekstraktlar hazırlanmıştır. Hazırlanan ekstraktların ilk olarak antimikrobiyal aktiviteleri ölçülmüştür.Antimikrobiyal aktivite için Triptic Soy Broth ve Mueller Hinton Agar laboratuvar ortamında hazırlanmış ve 121 °C'de 1 atm başınçta 15 dakika otoklavlanmıştır. Çıkarılan besiyeri steril edilmiş biyogüvenlik kabininde petrilere dökülmüş ve kullanılana kadar + 4 °C'de muhafaza edilmiştir. Hazırlanan besiyerlerine hazırlanan bakteri suşlarının ekimleri yapılmıştır. Daha sonra her bitki ekstraktının emdirildiği diskler petriler üzerine yerleştirilmiştir. Negatif kontrol olarak ekstraktların hazırlandığı çözücüler kullanılmıştır.Ekimi yapılan diskleri yerleştirilen petri kutuları 37 °C'de 24 saat inkübatör'de inkübasyona bırakılmış ve sonunda zon çapları ölçülmüştür. Antimikrobiyal aktivite 2 paralel olarak çalışılmış. Her bitkinin öncelikle tek başına antimikrobiyal etkisi ölçülmüş olup daha sonra birbirleri ile kombine edilerek antimikrobiyal etkilerine bakılmıştır ve sonuçlar birbirleri ile ve gentamicin ile karşılaştırılarak yorumlanmıştır. Antioksidan aktivite için hazırlanan ekstraktlardan 1 mL alınarak üzerine 1 mL %0,04'lük DPPH çözeltisi ilave edilerek karıştırılmıştır. Oda sıcaklığında ve karanlıkta bekletilen çözeltilerin absorbansları 517 nm'de ölçülmüştür. Çıkan değerlerden %50 inhibisyon sağlayan konsantrasyon değeri hesaplanmıştır. Antioksidan aktivite 3 paralel olacak şekilde çalışılmıştır. Elde edilen sonuçlar tek tek ve birbirleri ile karşılaştırılarak yorumlanmış en son askorbik asit ile karşılaştırılarak en yüksek antioksidan aktiviteye sahip ekstrakt belirlenmiştir. Toplam fenolik madde Folin-Ciocalteu reaktifi ile belirlenmiştir. Hazırlanan ekstraktlardan 100 μL alınarak 200 μL %50'lik Folin-Ciocalteu reaktifi eklenmiş ve 2 dakika bekletilmiştir. Üzerine 1 mL CO3 çözeltisi eklenerek 1 saat karanlıkta bekletilerek 760 nm'de absorbansları okunmuştur. Sonuçlar gallik asit cinsinden hesaplanıp, yorumlanmıştır. Yapılan çalışma sonucunda tüm değerler için tablolar oluşturulup, sonuçlar kendi içinde ve literatürde kayda geçen değerler ile karşılaştırılmış ve sonuçlar yorumlanmıştır. Bitkilerin birbiri ile kombine edilerek hazırlanan detoks suları üzerine literatürde çok fazla çalışma olmadığı için ,kombine olarak hazırlanan ekstraktlar sadece kendi içlerinde değerlendirip yorumlanmıştır. En yüksek antimikrobiyal aktivite limonda (Citrus limonum) ölçülürmüş olup, limon ile hazırlanan ekstrakt tüm bakteri suşları üzerinde yüksek aktivite göstermiştir. Maydanoz (Petroselinum crispum), salatalık (Cucumis sativa),zencefil (Zingiber officinale),ve avokado (Persea americana)'dan hazırlanan ekstraktların antimikrobiyal aktivite göstermediği belirlenmiştir. En yüksek antioksidan aktivite 30.47 μg/mL olarak elmada (Malus domestica) ölçülmüştür. Birbiri ile kombine edilerek hazırlana ekstraktlardan en yüksek antioksidan aktivite maydanoz-salatalık-limon ekstraktında ölçülmüştür. En yüksek antimikrobiyal aktivite maydanoz-limon ve limon- yeşil çay ekstraktlarında gözlemlenmiştir. En yüksek toplam fenolik madde içeriği ise limon-zencefilde ölçülmüştür. Çalışmada kullanılan bitkilerin tamamı yüksek antioksidan etkiye sahip olduğundan en düşük antioksidan aktivite değeri yüksek bulunmaktadır. Bitkilerin karışımları ile hazırlanan etanol ekstraktlarında limonun karıştırılması ile antimikrobiyal etkinliğin arttığı gözlemlenmiştir.Toplam fenolik madde miktarı tek bir bitkiden hazırlanan ekstraktlarda daha yüksek ölçülmüştür. Birbiri ile karıştırılarak hazırlanan ekstraktlarda toplam fenolik madde miktarında yükselme ölçülememiştir. Yapılan çalışmada kullanılan bitkilerin tek başına yüksek antioksidan özelliğe sahip olması sebebi ile, karışım olarak hazırlandığında antioksidan aktivite üzerinde herhangi bir değişime sebep olmadığı gözlemlenmiştir. Antimikrobiyal etkinliği karışım olarak hazırlanan ekstraktlarda limonun arttırdığı kaydedilmiştir. Çıkan sonuçlar doğrultusunda tüketilen detoks sularında yüksek antioksidan ve antimikrobiyal aktivite için limon ve elmanın karıştırılarak tüketilmesi önerilmektedir.Literatürde karışım olarak hazırlanan detoks sularının geniş mikrobiyolojik çalışmaları kısıtlı olduğundan çalışma ilk niteliği taşımaktadır.
Plants are as important organisms in nature as humans and animals. Mythologically, plants are the most valuable gift given to humans and animals. While plants are used as a food source, they have been preferred for centuries to eliminate health problems. Plants have been used by the people since ancient times for the removal of wounds and infections, as well as for the treatment of diseases. These therapeutic properties come from the fact that they contain some chemical substances that have a physiological effect on the human body.Some of these chemicals that give plants therapeutic properties are alkaloids, saponins, tannins, flavonoids, phenolic compounds. As the studies on plants have increased, the benefits of plants have come to light more and more.The important components contained in the content of plants increase plant consumption and reveal alternative ways of consumption. Detox is the elimination and neutralization of toxins present in our body, and there is a certain process for this.Plants help in this process. In our study, the activities of detox waters prepared with various plants were investigated. In the study, the total amount of phenolic substances, antioxidant and antimicrobial properties of mixtures prepared from some plants whose biological properties have been studied before were investigated. Study Malus domestica (apple), Zingiber officinale (ginger), Camellia sinensis (green tea), Cinnamomum (cinnamon), Citrus × limon (lemon), Petroselinum crispum (parsley), Cucumis sativus (cucumber), Persea americana Mill.( avocado), Spinacia oleracea (spinach) and ethanol extracts prepared by combining apple-cinnamon, lemon-ginger, lemon-green tea, parsley- lemon, salad-lemon, salad-parsley, avocado-lemon, parsley-salad-lemon these plants with each other B. subtilis ATCC 6633, S. aureus ATCC 29213, E. faecalis ATCC 29212, E. coli ATCC 25922, S. epidermidis ATCC 12228, S. abony NCTC 6017, S. typhimurium ATCC 14028 antimicrobial properties were investigated by disk diffusion method against the strains. Antioxidant effects and total phenolic substance amounts were determined by Folin Ciocalteu reagent using DPPH method. First, the fruits and vegetables bought from the transfer and Sunday were washed and left to brew in warm water. Detox waters prepared by brewing for about 1 hour were filtered with funnel and filter paper and blown away. The remaining substances in the blown detox water were dissolved with the ethanol concentration prepared to the appropriate extent and extracts from the plant detox waters were prepared.The antimicrobial activities of the prepared extracts were first measured. The extracted medium was poured into petri dishes in a sterilized biosafety cabinet and stored at + 4 °C until use. The prepared bacterial strains were October-ed in the prepared media. Then, the discs on which each plant extract was impregnated were placed on petri dishes.Solvents for the preparation of extracts were used as a negative control.The petri boxes with the October discs placed were incubated in the incubator at 37 °C for 24 hours and the zone diameters were measured at the end. Antimicrobial activity 2 was studied in parallel. The antimicrobial effect of each plant was first measured alone, then combined with each other to look at their antimicrobial effects, and the results were interpreted by comparing them with each other and with gentamicin. 1 mL of the extracts prepared for antioxidant activity was taken and mixed by adding 1 mL of 0.04% DPPH solution to it. The absorbances of the solutions kept at room temperature and in the dark were measured at 517 nm.The concentration value providing 50% inhibition was calculated from the values obtained. Antioxidant activity has been studied in 3 parallel ways. The obtained results were interpreted individually and compared with each other, the extract with the highest antioxidant activity was determined by comparing with the latest ascorbic acid. The prepared extracts were added to the tubes and 2.5 mL of phosphate buffer (0.2 M), 2.5 mL of 1% potassium ferrocyanide [K3Fe(CN6)] was added and mixed. Then the mixtures were left to rest for 30 minutes at 50 °C. At the end of the period, 2.5 mL of tri-chloroacetic acid (TCA) solution (10% in water) was added and centrifuged at 3000 rpm. 2.5 ml of the solution was taken from the supernatant part and 2.5 ml of distilled water and 0.5 ml of 0.1% iron chloride (FeCl3) were added. The absorbance values were read at 700 nm. The total phenolic substance was determined by Folin-Ciocalteu reagent.100 μL of the prepared extracts were taken and 200 μL of 50% Folin-Ciocalteu reagent was added and left for 2 minutes.The absorbance was read at 760 nm by adding 1 mL of CO3 solution to it and keeping it in the dark for 1 hour.The results were calculated in terms of gallic acid and interpreted. As a result of the study, tables were created for all values, the results were compared with the values recorded in the literature and the results were interpreted. Since there are not many studies in the literature on detox juices prepared by combining plants with each other, the extracts prepared in combination have only been evaluated and interpreted internally. The highest antimicrobial activity was measured in lemon (Citrus limonum), and the extract prepared with lemon showed high activity on all bacterial strains.Parsley (Petroselinum crispum), cucumber (Cucumis sativa), ginger (Zingiber officinale) and avocado (Persea americana) did not show antimicrobial activity. The highest antioxidant activity was measured in apples (Malus domestica) at 30.47 μg/mL. It has been observed that the extract prepared from spinach leaves has a high antimicrobial effect on all bacterial strains. It has been observed that the extract prepared by mixing parsley with lemon has a high antimicrobial effect on all bacterial strains.Lemon-green tea extract prepared by mixing green tea with lemon has a high antimicrobial effect. It was determined that while only green tea extract did not have antimicrobial activity on Salmonella abony, Salmonella typhimurium, Escherichia coli and Enterococcus faecalis bacteria, green tea-lemon mixture exhibited activity. The highest antioxidant activity of the extracts prepared by combining with each other was measured in parsley-salad-lemon extract. The highest antimicrobial activity was observed in parsley-lemon and lemon- green tea extracts. The highest total phenolic substance content was measured in lemon-ginger. Among the extracts prepared as cinnamon, apple and apple cinnamon, the highest iron reducing power capacity was measured in the extract prepared from cinnamon, followed by apple. It was observed that the iron reducing power capacity decreased in the extract prepared with the combination of apple and cinnamon. In the prepared extracts, the extract consisting of lemon and green tea has a higher effect than spinach, and the values fluctuate depending on the concentration. The highest phenolic content among the extracts prepared by combining the herbs and plants we used in our study was measured in the detox water extract prepared with plain green tea. The phenolic content of detox water prepared with lemon and ginger was found to be close to green tea, and the least phenolic content was measured in detox waters prepared with avocado, apple and avocado-lemon mixture. Since all of the plants used in the study have a high antioxidant effect, the lowest antioxidant activity value is high.It has been observed that the antimicrobial activity increases with the mixing of lemon in ethanol extracts prepared with mixtures of plants. The total amount of phenolic substances was measured higher in extracts prepared from a single plant. An increase in the total amount of phenolic substances in extracts prepared by mixing with each other could not be measured. Due to the fact that the plants used in the study have high antioxidant properties alone, it has been observed that when prepared as a mixture, it does not cause any change in antioxidant activity.It has been recorded that lemon increases the antimicrobial activity in extracts prepared as a mixture. According to the results obtained, it is recommended to consume lemon and apple by mixing for high antioxidant and antimicrobial activity in the detox waters consumed. Since extensive microbiological studies of detox waters prepared as a mixture are limited in the literature, the study is of a preliminary nature.
