Bazı steroidlerin mucor hiemalis küfü ile biyotransformasyonları

Abstract

Bu çalışmada, pregnenolon ve dehidroepiandrosteron (DHEA) steroidlerinin Mucor hiemalis MRC 70325 küfünde nasıl metabolize edileceklerini incelemek için bu steroidlerin söz konusu küf ile beş gün süren biyotransformasyonları gerçekleştirildi. Pregnenolon bileşiğinin M. hiemalis MRC 70325A ile inkübasyonundan 3ß,7?-dihidroksipregn-5-en-20-on (% 64), 3ß,7?,11?-trihidroksipregn-5-en-20-on (% 11) ve yeni bir metabolit olan 3ß,14?-dihidroksipregn-5-en-7,20-dion (%3) elde edildi. Dehidroepiandrosteron bileşiğinin M. hiemalis MRC 70325 ile inkübasyonundan ise 3ß,17ß-dihidroksiandrost-5-en (%6) ve 3ß,7?-dihidroksiandrost-5-en-17-on (%72) metabolitleri elde edildi. Metabolitlerin yapı tayinleri, başlangıç maddeleri ile metabolitlerin erime noktaları, NMR ve IR spektrumları karşılaştırılarak gerçekleştirildi.

In this work, pregnenolone and dehydroepiandrosterone (DHEA) were incubated by Mucor hiemalis MRC 70325 for 5 days in order to see how these steroids would be metabolized by the fungus. Incubation of pregnenolone with M. hiemalis MRC 70325 afforded 3ß,7?-dihydroxypregn-5-en-20-one (64%), 3ß,7?,11?-trihydroxypregn-5-en-20-one (11%) and 3ß,14?-dihydroxypregn-5-en-7,20-dione (3%). 3ß,14?-Dihydroxypregn-5-en-7,20-dione was determined as a new metabolite. Incubation of dehydroepiandrosterone with M. hiemalis MRC 70325 afforded 3ß,17ß-dihydroxyandrost-5-ene (6%) and 3ß,7?-dihydroxyandrost-5-en-17-one (72%). The structures of the metabolites were elucidated by comparing melting points, NMR and IR spectra of the starting materials with those of metabolites.
