Bu çalışmada, ilaç hammaddesi olma potansiyeli düşünülerek Sakarya Üniversitesi Kimya Bölümü'nde yeni sentezlenen ve insan karbonik anhidraz I ve II izoenzimlerini inhibe ettiği belirlenen ftalazin içerikli β-Laktam türevi (IV-b)'nin genotoksik ve sitotoksik profillerinin belirlenmesi amaçlanmıştır. İlaç hammaddesi olarak sunulması düşünülen kimyasal maddeler çok çeşitli toksikolojik araştırmalardan geçirilir. Bu araştırmalardan bir kısmı da sitotoksisite ve genotoksisite çalışmalarıdır. Bu bağlamda in vitro insan periferal kan lenfositleri üzerine test maddesi IV-b'nin sitotoksik ve genotoksik etkilerinin belirlenmesi için kromozomal anormallik (CA) ve mikronukleus (MN) testleri uygulanmıştır. Bu amaçla lenfosit kültürü 24 ve 48 saat süre ile test maddesinin dört farklı konsantrasyonu (3,75, 7,5, 15, 30 µg/mL) ile muamele edilmiştir. Araştırmada elde edilen verilere göre, test maddesinin bütün uygulama gruplarında negatif kontrole göre mitotik indeksi anlamlı bir şekilde azalttığı gözlenmiştir. Çözücü kontrole göre ise, 24 saatlik uygulamada sadece 3,75 µg/mL'lik konsantrasyon ile 48 saatlik uygulamada 7,5 ve 15 µg/mL'lik konsantrasyonlarda anlamlı azalışlar gözlemlenmiştir. Bütün uygulama gruplarında en fazla gözlemlenen kromozomal anormallik tipi kromatid kırığı iken, bunu fragment ve kromozom kırıkları izlemiştir. Anormal hücre yüzdesi ve hücre başına düşen kromozomal anormallikler bakımından negatif kontrole göre her iki uygulama süresinde de en yüksek iki konsantrasyonda anlamlı artışlar gözlenmiştir. Ayrıca 24 saatlik uygulamada 7,5 µg/mL'lik konsantrasyonda da anlamlı artış söz konusudur. Her iki değerlendirme kriteri açısından her iki uygulama süresinde çözücü kontrole göre en yüksek konsantrasyonda anlamlı artışlar görülmüştür. Bununla birlikte hücre başına düşen kromozomal anormallikler bakımından 48 saalik uygulamada 15 µg/mL'lik konsantrasyonda da anlamlı bir artış görülmüştür. Mikronukleus frekansı bakımından, en düşük konsantrasyon hariç bütün konsantrasyonlarda çözücü kontrole göre anlamlı artışlar söz konusudur. Elde edilen sonuçlara göre, test maddesinin uygulanan konsantrasyonlarının in vitro şartlarda sitotoksik etkili olduğu, özellikle yüksek konsantrasyonlarda ise genotoksik etkili olduğu görülmüştür.
The aim of this study was to determine the genotoxic and cytotoxic profile of the phthalazine-containing β-lactam derivative (IV-b), which is newly synthesized in Sakarya University Chemistry Department and inhibited the isoenzymes of human carbonic anhydrase I and II. Chemical substances which are considered as pharmaceutical raw materials are subjected to a wide variety of toxicological researches. Some of these studies are cytotoxicity and genotoxicity studies. In this context, chromosomal abnormality (CA) and micronucleus (MN) tests were performed to determine the cytotoxic and genotoxic effects of test substance IV-b on in vitro human peripheral blood lymphocytes. For this purpose, the lymphocyte culture was treated with four different concentrations of test substance (3,75, 7,5, 15, 30 µg/mL) for 24 and 48 hours. According to the data obtained in the study, it was observed that the test substance significantly reduced the mitotic index in all application groups compared to the negative control. According to the solvent control, significant reduces were observed in the concentration of 7,5 and 15 µg/mL in 48 hours with only 3,75 µg/mL concentration in 24 hours of application. The most observed chromosomal abnormality was chromatid fracture in all groups, followed by fragment and chromosome fractures. Significant increases in the maximum two concentrations were observed with respect to the percentage of abnormal cells and chromosomal abnormalities per cell. There is also a significant increase in the concentration of 7,5 µg/mL in 24 hours. For both evaluation criteria, significant increases were observed in the highest concentration of solvent control in both application periods. However, in terms of chromosomal abnormalities per cell, a significant increase in the concentration of 15 µg/mL was observed in 48 hours. In terms of micronucleus frequency, there is a significant increase in all concentrations except the lowest concentration compared to solvent control. According to the results obtained, the applied concentrations of the test substance were cytotoxic in in vitro conditions and genotoxic effect, especially at high concentrations.
