Rapeseed (Brassica napus ssp. oleifera L.), from Crucifera family, has been widely used as an oil crop all over the world. The application of biotechnology to crop improvement requires efficient adventitious plant regeneration protocols. The introduction of foreign genes coding for agronomically important traits into plant cells is useless unless transgenic plants are produced. The adventitious shoot regeneration capacity of cells or tissues is the main factor affecting the success of all transformation techniques. In the present study aiming to establish an efficient regeneration protocol in rapeseed, the effects of different growth mediums (MS and B5) and plant growth regulators (6-benzylaminopurine (BAP), naphthaleneacetic acid (NAA) and thidiazuron (TDZ)) at different concentrations on the regeneration capacity of hypocotyl and stem explants of three rapeseed cultivars (‘Spok’, ‘Westar’ and ‘Helios’) were investigated. The highest values were obtained from the stem explant cultured on MS medium containing 3 mg l−1 BAP and 0.2 mg l−1 NAA. Among three cultivars, ‘Spok’ showed the best performance with respect to tissue culture response. |
